But how does an ingestible painkiller actually work so that your head can clear half an hour later?
Once swallowed, the pill or liquid solution travels through the body and is absorbed into the bloodstream. At that point, the blood carries the medicine to different parts of the body, looking for pain.

Ibuprofen (Advil), naproxen (Aleve), and aspirin are part of a class of drugs called nonsteroidal anti-inflammatory drugs. They treat pain by reducing inflammation (which can be anything from heat, fever, swelling, pain, or loss of movement). Say you strain your back muscles while moving into a new apartment: The body’s white blood cells go there to repair the muscles, causing swelling and heat that can cause pain. (This is one reason why a little pain can be good: Your body is saying: I’m hurt, take it easy on me.)
On a chemical level, as noted in a 2012 TED video, aspirin and ibuprofen (along with naproxen) work by binding to an enzyme called cyclooxygenase, blocking it. This prevents the body from producing molecules called prostaglandins, which generate inflammation that often leads to pain. With the molecule blocked, the pain begins to subside.
Although Tylenol has been on the market since the 1950s, we still don’t have a good idea of how it works. As one researcher told Carmen Drahl, for an article in C&E News, “It’s not an opioid and it’s not an NSAID… The question is, what is it?”
Although there are many theories (for example, it may have a lot to do with inhibiting the same enzymes as NSAIDs), what we do know is that it is used to treat pain and fever. Unlike NSAIDs, it does not treat inflammation.
